Towards the Single Use of TFF in ADC Manufacturing

Tangential Flow Filtration (TFF) is a common unit operation in ADC manufacturing and enables concentration and exchange to pre-formulation buffer. The presence of toxic linker-payloads following conjugation presents challenges to traditional TFF operation that can be addressed by single use processes. In this presentation, the performance, ease of use and safety of a novel single-use TFF capsule is evaluated and compared to conventional Pellicon® 3 TFF cassettes.

To ensure successful transfer from lab scale to GMP production, robust, scalable platforms are essential. Single-use technologies provide many advantages, including improved safety, lower costs and greater flexibility. This poster will outline the advantages of a single-use platform and demonstrate scalability in the production of ADCs from development to GMP manufacturing.
