Application NotesAffinity Purification (use Cat. No. NA02A, see application references)
Immunocytochemistry (0.5-5 µg/ml)
Immunoprecipitation (1-2 µg, see application references)
Paraffin Sections (see application references)
Application CommentsAlso recognizes 7-methylguanosine-capped mRNA. This antibody reacts with the exposed 5' cap structure of the snRNAs and the antibody can be used for the purification of snRNPs by immunoaffinity chromatography (use Cat. No. NA02A). All of the major snRNPs bind to the monoclonal antibody column and can be gently eluted by washing with 7-methyl guanosine. Antibody should be titrated for optimal results in individual systems.

DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with NS-1 mouse myeloma cells. Recognizes the exposed 5′ cap structure (2,2,7-trimethylguanosine) of small nuclear RNAs (snRNAs).
BackgroundThe U small nuclear ribonucleoprotein particles (snRNPs) are present in all eukaryotes studied thus far and are involved in the splicing of RNA molecules. snRNPs are composed of several proteins and one or two RNA molecules. The RNA molecules (snRNAs) contain a unique 5' terminal cap structure containing the nucleoside 2,2,7-trimethylguanosine. The 70 kDa protein component of U1 snRNP is cleaved during apoptosis, perhaps by a caspase. Monoclonal antibodies to the 5' cap structure react only to snRNAs.
