Background InformationHepatocyte growth factor (UniProt: P14210; also known as Hepatopoietin-A, Scatter factor, SF, HGF) is encoded by the HGF (also known as HPTA) gene (Gene ID: 3082) in human. HGF is secreted glycoprotein that is synthesized with a signal peptide (aa 1-31) that is subsequently cleaved off to generate the mature form that contains an - chain (aa 32-494) and a β-chain (495-728) that are linked by a disulfide bridge. Newly synthesized HGF is a single-chain inactive protein (scHGF) that is proteolytically processed by extracellular proteases between arginine 494 and valine 495 into two-chain HGF (tcHGF). Only the tcHGF has biological activity and activates the MET receptor. HGF contains four Kringle domains (aa 128-206, 211-288, 305-383, and 391-465) and one serine protease (SP) domain (495-721). Clone t8E4 is reported to recognize human HGF and does not recognize mouse HGF. It selectively binds only to tcHGF and not to scHGF. It recognizes an epitope present in the K4-SP region, and that this site is formed only after the cleavage between arginine 494 and valine 495.
