Background InformationPackaging protein 3 (UniProt: P04496; also known as L1-52/55 kDa protein, Packaging protein 52K) is encoded by the L1 gene (Gene ID: 2652993) in human Adenovirus 5. Adenovirus 5 belongs to the Adenoviridae family of non-enveloped viruses that are characterized by linear, double-stranded DNA genomes. Their capsid is an icosahedron that contains at least 12 different viral proteins. Adenovirus 5 is commonly used for gene transfer experiments due to its ability to infect a wide group of different cell types. It can harbor large genes in its genome incorporated via homologous recombination techniques. The 52 kDa packaging protein of human adenovirus 5 is part of the genome packaging machinery that consists of polypeptides IIIa, IVa2, L4 33k, and L4 22k. It is expressed in the early phase and late phase of viral replicative cycle. Its role during assembly consists in tethering the viral core to the icosahedral shell wherein maturation proceeds simultaneously with packaging, before the viral particle is sealed. Its adenovirus protease consensus cleavage site is localized at amino acids 351-352. It plays a critical role in the coordinated assembly of infectious progeny particles and contributes to the specificity of packaging, possibly by interacting with capsid protein IIIa. It is proteolytically cleaved at different sites during virion maturation. It is reported to be present in empty particles in its full-length form but is absent from the mature virion.
