Background InformationCadherin-17 (UniProt: Q12864; also known as Intestinal peptide-associated transporter HPT-1, Liver-intestine cadherin, LI-cadherin) is encoded by the CDH17 gene (Gene ID: 1015) in human. Cadherin-17 is a single-pass type I membrane protein that is expressed in the gastrointestinal tract and pancreatic duct. It belongs to the non-classical cadherin with distinct structural feature of having seven cadherin repeats at its extracellular amino-terminus. Three calcium ions are usually bound at the interface of each cadherin repeat and rigidify the connections, imparting a strong curvature to the full-length ectodomain. Cadherin-17 is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature form that contains a long extracellular domain (aa 23-787), a transmembrane domain (aa 788-808), and a cytoplasmic domain (aa 809-832). It differs from classical cadherins in having a short cytoplasmic tail at its C-terminus. Cadherin-17 is reported to be a oncofetal protein that is associated with poor prognostic outcome in subjects with hepatocellular carcinoma. Its levels can be up-regulated or down-regulated in different cancers, including pancreatic, gastric and colorectal cancers. Clone Lic5 is shown to reduce the levels of beta-catenin and in hepatocellular carcinoma cells and administration of Lic5 is reported to induce the activation of caspase-8 and 9 in these cells. In addition, it is also shown to suppress tumor growth of MHCC97L cells in murine models. (Ref.: Wang, Y., et al. (2013). PLoS One. 8(8); e72386).
