Background InformationCaspase-1, also known as IL-1 beta-converting enzyme (ICE), is an ezyme that cleaves the inflammatory cytokines IL-1 beta and IL-18, releasing the mature, active peptides. Caspase-1 is important for defense against pathogens and it also is involved in the induction of apoptosis. Thymocytes from caspase-1 deficient mice are resistant to Fas-mediated apoptosis, and caspase-1 is similar to the cell death gene CED-3 of C. elegans. Caspase-1 is a heterotetramer that consists of two anti-parallel heterodimers, each one formed by a 20 kDa (p20) and a 10 kDa (p10) subunit. The enzyme has a preferred cleavage site of Tyr-Val-Ala-Asp (YVAD). Caspase-1 is highly expressed in spleen and lung, and is absent from brain.

Entrez Gene Number
Entrez Gene SummaryThis gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce 2 subunits, large and small, that dimerize to form the active enzyme. This gene was identified by its ability to proteolytically cleave and activate the inactive precursor of interleukin-1, a cytokine involved in the processes such as inflammation, septic shock, and wound healing. This gene has been shown to induce cell apoptosis and may function in various developmental stages. Studies of a similar gene in mouse suggest a role in the pathogenesis of Huntington disease. Alternative splicing of this gene results in five transcript variants encoding distinct isoforms.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P29466 # Thiol protease that cleaves IL-1 beta between an Asp and an Ala, releasing the mature cytokine which is involved in a variety of inflammatory processes. Important for defense against pathogens. Cleaves and activates sterol regulatory element binding proteins (SREBPs). Can also promote apoptosis.
SIZE: 404 amino acids; 45159 Da
SUBUNIT: Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 20 kDa (p20) and a 10 kDa (p10) subunit. The p20 subunit can also form a heterodimer with the epsilon isoform which then has an inhibitory effect. May be a component of the inflammasome, a protein complex which also includes PYCARD, CARD8 and NALP2 and whose function would be the activation of proinflammatory caspases.
SUBCELLULAR LOCATION: Cytoplasm.
TISSUE SPECIFICITY: Expressed in larger amounts in spleen and lung. Detected in liver, heart, small intestine, colon, thymus, prostate, skeletal muscle, peripheral blood leukocytes, kidney and testis. No expression in the brain.
PTM: The two subunits are derived from the precursor sequence by an autocatalytic mechanism.
SIMILARITY: SwissProt: P29466 ## Belongs to the peptidase C14 family. & Contains 1 CARD domain. ... hide » see more »

Product Usage Statements
Quality AssuranceRoutinely evaluated by Western Blot on cell lysate from human HL-60.

Western Blot Analysis:
0.5-2 μg/mL of this lot of antibody detected the p45 proenzyme form of Caspase 1 in 20 μg of cell lysate from human HL-60. The immuno-reactivity can be inhibited by the immunizing peptide.
Usage Statement
  • Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage ConditionsStable for 1 year at -20ºC from date of receipt.

Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol-containing solutions to become frozen during storage.

FAQ

QuestionAnswer
Why does caspase often exhibit different molecular weights?Full-length caspase 3 (pro-form) is 32kDa. Upon activation, caspase-3 is cleaved generating two smaller subunits of 17 kDa and 12 kDa. Other Caspases include Caspase 1 (proenzyme at 45kDa and subunit at 20kDa); Caspase 6 at 34kDa; Caspase 7 at 38kDa; Caspase 8 at 55kDa and Caspase 9 at 46-48kDa. It should be noted however that it can be difficult to detect the active fragments of many caspases because of their small size and biologically short life. The pro-forms are much more stable and readily detectable.
