Background InformationChromatin licensing and DNA replication factor 1, also known as CDT1, is a key licensing factor which, along with the protein Cdc6, functions to license DNA by forming the pre-replicative complex (pre-RC). Its activity during the cell cycle is tightly regulated by its association with the protein geminin, which both inhibits Cdt1 activity during S phase in order to prevent re-replication of DNA and prevents it from ubiquitination and subsequent proteolysis.

SUBUNIT STRUCTURE: Interaction with GMNN inhibits both binding of the MCM complex to origins of replication and DNA-binding activity.
SUBCELLUAR LOCATION: Nucleus.
DEVELOPMENTAL STAGE: Present during G1 and early S phase of the cell cycle. Degraded during the late S, G2, and M phases.
PTM: Ubiquitinated, leading to its subsequent degradation. The interaction with GMNN protects it against ubiquitination.
Phosphorylated by cyclin A-dependent kinases which results in the binding of CDT1 to the F-box protein SKP2 and subsequent degradation. Binding to GMNN is not affected by phosphorylation.
