Background InformationCentrosomal protein of 55 kDa (UniProt: Q53EZ4; also known as Cep55, Upregulated in colon cancer 6) is encoded by the CEP55 (also known as C10orf3, URCC6) gene (Gene ID: 55165) in human. Cep55 is a homodimeric protein that plays a role in mitotic exit and successful completion of cytokinesis. It is located in the centrosome in interphase cells and recruited to the midbody during cytokinesis. Cep55 is highly expressed in testis and its expression is also reported in fetal brain ganglionic eminence, kidney tubules, and multinucleate neurons in the temporal cortex. Its expression has also been reported in several cancer cell lines. Cep55 is present at the centrosomes at interphase. A small portion is associated preferentially with the mother centriole, whereas most of it localizes to the pericentriolar material. During mitosis, it loses its affinity for the centrosome at the onset of prophase and diffuses throughout the cell. This dissociation from the centrosome is phosphorylation dependent. In Cep55, serine 425 and 428 are shown to be phosphorylated at the onset of mitosis and prior to serine 436 phosphorylation. These phosphorylations are essential for dissociation from the centrosome at the G2/M boundary. In Western blot analysis of synchronized cells, Cep55 may appear as a doublet that is indicative of phosphorylation during mitosis, and a single protein in interphase cells. In cancer cells, Cep55 expression is correlated with cell growth rate and p53 is shown to negatively regulate its levels and promoter activity. Down-regulation of Cep55 is accompanied by repression of polo-like kinase 1 (PLK1). Over-expression of PLK1 and knockdown of p53 are reported to enhance the post-translational stability of Cep55.
