Background InformationUbiquitin-like with PHD and ring finger domains 1 (UHRF1), also known as ICBP90 or Np95, is a multifunctional nuclear protein that may be an important mediator of heterochromatin formation which is indicative of gene repression. UHRF1 contains a set and RING-finger-associated domain that detects hemi-methylated cytosine residues leading to the recruitment of DNMT1 to these residues during the S phase. UHRF1 thereby facilitates the transfer of methylation status to newly synthesized DNA molecules. UHRF1 may also interact with histones and HDAC1 via its PHD domain to promote modifications of histone residues which contribute to heterochromatin formation. UHRF1 may also directly modify histone H3 by ubiquitination. In addition, UHRF1 may contribute to the DNA-repair process though associations with PCNA. Several studies have suggested that overexpression of UHRF1 may contribute to the development of many cancers.

Entrez Gene Number
Entrez Gene SummaryThis gene encodes a member of a subfamily of RING-finger type E3 ubiquitin ligases. The protein binds to specific DNA sequences, and recruits a histone deacetylase to regulate gene expression. Its expression peaks at late G1 phase and continues during G2 and M phases of the cell cycle. It plays a major role in the G1/S transition by regulating topoisomerase IIalpha and retinoblastoma gene expression, and functions in the p53-dependent DNA damage checkpoint. Multiple transcript variants encoding different isoforms have been found for this gene.
Gene Symbol
  • UHRF1
  • ICBP90
  • NP95
  • RNF106
Purification MethodUnpurified
UniProt Number
UniProt SummaryFUNCTION: Putative E3 ubiquitin-protein ligase. May participate in methylation-dependent transcriptional regulation. Binds to inverted 5'-CCAAT-3' box 2 in the TOP2A promoter, and activates TOP2A expression. Important for G1/S transition. May be involved in DNA repair and chromosomal stability.

PATHWAY: Protein modification; protein ubiquitination.

SUBUNIT STRUCTURE: Interacts with histones H3, H1 and H2B. Interacts with HDAC1, but not with HDAC2. Interacts with UHRF1BP1. Binds methylated CpG containing oligonucleotides.

SUBCELLULAR LOCATION: Nucleus.

TISSUE SPECIFICITY: Expressed in thymus, bone marrow, testis, lung and heart. Overexpressed in breast cancer.

DEVELOPMENTAL STAGE. Expressed in fetal thymus, liver and kidney.

INDUCTION: Up-regulated in proliferating cells, and down-regulated in quiescent cells. Down-regulated upon adriamycin-induced DNA damage, in a p53/TP53 and CDKN1A-dependent way. Induced by E2F1 transcription factor.

DOMAIN:The RING finger is required for ubiquitin ligase activity. The YDG domain mediates the interaction with histone H3

POST-TRANSLATIONAL MODIFICATION: Phosphorylated on serine residues. Phosphorylation may enhance DNA-binding activity. Ubiquitinated; which leads to proteasomal degradation. Polyubiquitination may be stimulated by DNA damage.

SEQUENCE SIMILARITIES: Contains 1 PHD-type zinc finger. Contains 2 RING-type zinc fingers. Contains 1 ubiquitin-like domain. Contains 1 YDG domain.

SEQUENCE CAUTION: The sequence BAB15177.1 differs from that shown. Reason: Erroneous initiation.
