Background InformationIntegrin beta-7 (UniProt: P26011; also known as Integrin beta-P, M290 IEL antigen) is encoded by the Itbg7 gene (Gene ID: 16421) in murine species. Integrins are heterodimeric integral membrane proteins composed of a variable -subunit and a conserved β-subunit that function in cell surface adhesion and signaling. They contain a large extracellular domain responsible for ligand binding, a single transmembrane domain, and a cytoplasmic domain. The exact combination of various and β-subunits dictates the binding specificity of integrins to different ECM components. Although both subunits are required for adhesion, the binding specificity primarily depends on the extracellular portion of the -subunit. Integrin β7 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to produce the mature form that contains a large cytoplasmic domain (aa 20-724), a transmembrane domain (aa 725-745), and a cytoplasmic domain (aa 746-806). It associates with associates with either 4 or E (ITGAE). Eβ7 can then function as a receptor for E-cadherin. 4β7 is involved in adhesive interactions of leukocytes and serves as a receptor for fibronectin. It recognizes one more domains within the alternatively spliced CS-1 region of fibronectin. It can also serve as receptor for MAdCAM1 and VCAM1 and is reported to recognize the sequence L-D-T in MAdCAM1. Shp1 is shown to selectively inhibit endocytosis of β7, which enhances surface 4β7 display and lymphocyte homing to gut-associated lymphoid tissue. Shp1 is reported to restrain plasma membrane β7 phosphorylation and inhibit β7 endocytosis. Clone FIB504 is reported to functionally block human 4β7. (Ref.: Ballet, R., et al. (2021). Nat.
