Background InformationNeuroligin-4 (UniProt: Q8NFZ3; also known as Neuroligin Y) is encoded by the NLGN4Y (also known as KIAA0951) gene (Gene ID: 22829) in human. Neuroligins are postsynaptic cell adhesion molecules that have crucial roles in synapse maturation and function. In humans, NLGN4X is located on the X chromosome and the NLGN4Y is localized on the Y chromosome and together they form an X-Y pair. They share about 97% amino acid identity; however, they are differentially regulated. Although several mutations in NLGN4X have been linked to autism, only one mutation has been identified in NLGN4Y gene. Despite their high degree of sequence identity, NLGN4Y exhibits severe deficit in trafficking to cell surface and related synaptic effects. This has been attributed to serine 93 in NLGN4Y. A substitution of serine to proline at residue 93 is shown to rescue it and increase its surface trafficking. Neuroligin-4 is a single-pass type I, homodimeric, membrane glycoprotein that is synthesized with a signal peptide (aa 1-43), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 44-676), a transmembrane domain (aa 677-697), and a cytoplasmic domain (aa 698-816). Its expression is observed in fetal and adult brain, prostate, and testis. It serves as a neuronal cell surface protein that is involved in cell-cell interactions. (Ref.: Nguyen, TA., et al. (2020).
