Background InformationOligodendrocytes are large glial cells found in the central nervous system. They are responsible for production of myelin sheath that insulates neuronal axons. The first step in the generation of oligodendrocytes is the emergence of oligodendrocyte precursors (OPCs) from embryonic radial glia. Subsequently, OPCs differentiate into immature oligodendrocytes before they begin to generate myelin. This process is regulated by both intrinsic and extrinsic signals in cells. Oligodendrocytes have only a brief period for myelination early during differentiation, and mature oligodendrocytes are incapable of myelinating. An essential signal for the onset of myelination is reported to be provided by the electrical activity of neurons. Although the ATP released from axons firing axon potentials does not directly act on OPCs or oligodendrocytes, it triggers the release of leukemia inhibitory factor (LIF) from astrocytes, which promotes myelination. Although the electrical activity of neurons is an essential promyelinating factor, other factors also come into play in this process. During development, all growing nerve fibers are shown to express the polysialylated-neural cell adhesion molecule (PSA-NCAM) that can prevent homophilic NCAM adhesion and cell-cell interactions. When this form of NCAM is downregulated, as it is in electrically active neurons, only then the process of myelination can proceed. (Ref.: Bradl, M., and Lassmann, H. (2010).
