DescriptionAnti-RAD18 Antibody
Alternate Names
  • E3 ubiquitin-protein ligase RAD18
  • Postreplication repair protein RAD18
  • hHR18
  • hRAD18
  • RING finger protein 73
Background InformationHuman Rad18 (hRad18) is a homologue of the S. pombe Rad18. In yeast, Rad6 and Rad18 play a crucial role in post−replication repair functions in gap−filling of daughter strands on replication of damaged DNA. Furthermore, Rad6−Rad18 interaction is thought to be critical at an early step during lesion bypass in yeast. In vivo, Rad18 protein forms a tight complex with Rad6 through a conserved ring−finger motif. It has been suggested that the Rad18−Rad6 complex binds to the damaged sites by the single−stranded DNA binding property of Rad 18. This is followed by degradation of the stalled components of DNA replication by the ubiquitin−conjugating enzyme activity of Rad6. The human Rad18 maps on chromosome 3p24−25, the region which is often deleted in lung, breast, ovary, and testis cancers. The human RAD18 gene codes for a protein of 495 amino acids with a predicted molecular weight of 56 kDa. The human Rad18 is ubiquitously expressed in various human tissues.

Applications
ApplicationCat. No. AB1378, Anti-RAD18 Antibody is a rabbit polyclonal antibody against human RAD18 for use in Immunocytochemistry, Immunoprecipitation, and Western Blotting.
Key Applications
  • Western Blotting
  • Immunocytochemistry
  • Immunoprecipitation
Application NotesImmunocytochemistry Analysis: A representative lot detected both nuclear and cytosolic hRad18 by fluorescent immunocytochemistry staining of 3.7% formaldehyde- and 80% methanol-fixed GM637 human fibroblasts following DNA damage induction by UV irradiation. Prior 0.1% NP-40 treatment greatly reduced cytosolic staining (Miyase, S., et al. (2005). J. Biol. Chem. 280(1):515-524).
Immunocytochemistry Analysis: A representative lot detected increased human Rad18 (hRad18) nuclear foci co-localized with PCNA at the replication stalling sites by fluorescent immunocytochemistry staining of methanol-fixed GM637 human fibroblasts following DNA damage induction by UV irradiation (Watanabe, K., et al. (2004). EMBO J. 23(19):3886–3896).
Immunocytochemistry Analysis: A representative lot detected predominant nuclear localization of exogenously expressed human Rad18 (hRad18) by fluorescent immunocytochemistry staining of 3.7% formaldehyde- and 80% methanol-fixed primary p53-/- mouse fibroblasts microinjected with either wild-type or C207F mutant hRad18-expressing plasmid. Exogenously expressed C28F mutant hRad18 was seen with both cytosolic and nuclear localization, but became predominantly nuclear following DNA damage induction by UV irradiation (Tateishi, S., et al. (2000). Proc. Natl. Acad. Sci. U. S. A. 97(14):7927–7932).
Immunoprecipitation Analysis: A representative lot immunoprecipitated both non-ubiquitinated and ubiquitinated hRad18 from various human cell lysates as well as from in vitro ubiquitination reactions (Miyase, S., et al. (2005). J. Biol. Chem. 280(1):515-524).
Immunoprecipitation Analysis: A representative lot co-immunoprecipitated exogenously expressed polymerase η (polη), but not polδ, with hRad18 from lysates of transfected GM637 human fibroblasts exposed to UV irradiation (Watanabe, K., et al. (2004). EMBO J. 23(19):3886–3896).
Western Blotting Analysis: A representative lot detected both non-ubiquitinated and monoubiquitinated hRad18 in multiple human cell lysates. The monoubiquitinated hRad18 was detected predominantly in the cytosolic, but not nuclear, fraction from UV-irradiated GM637 human fibroblasts (Miyase, S., et al. (2005). J. Biol. Chem. 280(1):515-524).
Western Blotting Analysis: A representative lot detected exogenously expressed FLAG-tagged hRad18 with either full-length sequence or R6BD (Rad6-binding domain; a.a. 340-395) deletion in anti-FLAG immunoprecipitate from transfected cells, while only full-length hRad18, but not R6BD hRad18 co-immunoprecipitated with T7-tagged hRad6A/B in anti-T7 immunoprecipitate (Watanabe, K., et al. (2004). EMBO J. 23(19):3886–38964).
