DescriptionAnti-REST Antibody
Alternate Names
  • Neural-restrictive silencer factor
  • RE1-silencing transcription factor
  • X2 box repressor
  • neuron restrictive silencer factor
  • repressor binding to the X2 box
Background InformationRE1-silencing transcription factor (UniProt: Q13127; also known as REST, Neural-restrictive silencer factor, X2 box repressor) is encoded by the REST (also known as NRSF, XBR) gene (Gene ID: 5978) in human. REST is a transcriptional repressor that acts by binding a DNA sequence element called the neuron-restrictive silencer element (NRSE). It is ubiquitously expressed, and higher expression is found in the tissues of the lymphocytic compartment, including spleen, thymus, peripheral blood lymphocytes, and ovary. It is also found in undifferentiated neuronal progenitor cells and is believed to acts as a master negative regular of neurogenesis. Expression of REST is reported to be cell cycle-dependent with reduced levels in the G2 phase. It is involved in maintaining the quiescent state of adult neural stem cells and prevents their premature differentiation into mature neurons. REST restricts the expression of neuronal genes by associating with two distinct corepressors, SIN3A and RCOR1, which in turn recruit histone deacetylase to the promoters of REST-regulated genes. REST also mediates repression by recruiting the BHC complex at RE1/NRSE sites, which acts by deacetylating and demethylating specific sites on histones and acting as a chromatin modifier. Mutations in REST gene can lead to Wilms tumors, a pediatric malignancy of the kidney that occurs due to uncontrolled multiplication of renal stem, stromal, and epithelial cells. Applications
ApplicationAnti-REST, Cat. No. 09-019, is a rabbit polyclonal antibody that detects RE1-silencing transcription factor (REST) and is tested for use in Chromatin Immunoprecipitation (ChIP) and Western Blotting.
Key Applications
  • Western Blotting
  • Chromatin Immunoprecipitation (ChIP)
Application NotesTested Applications

Chromatin Immunoprecipitation Analysis (ChIP): 2 µg from a representative lot detected REST in Chromatin prepared from sonicated HeLa S3 cells (4 X 106 cell equivalents per IP).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Entrez Gene Number
Entrez Gene SummaryThis gene encodes a transcriptional repressor which represses neuronal genes in non-neuronal tissues. It is a member of the Kruppel-type zinc finger transcription factor family. It represses transcription by binding a DNA sequence element called the neuron-restrictive silencer element. The protein is also found in undifferentiated neuronal progenitor cells, and it is thought that this repressor may act as a master negative regular of neurogenesis. Alternatively spliced transcript variants have been described; however, their full length nature has not been determined. [provided by RefSeq]

UniProt Number
UniProt SummaryFunction: Transcriptional repressor which binds neuron-restrictive silencer element (NRSE) and represses neuronal gene transcription in non-neuronal cells. Restricts the expression of neuronal genes by associating with two distinct corepressors, mSin3 and CoREST, which in turn recruit histone deacetylase to the promoters of REST-regulated genes. Mediates repression by recruiting the BHC complex at RE1/NRSE sites which acts by deacetylating and demethylating specific sites on histones, thereby acting as a chromatin modifier.
Subunit structure: Interacts with SIN3A, SIN3B and RCOR1.
Subcellular location: NucleusProbable.
Tissue specificity: Ubiquitous. Expressed at higher levels in the tissues of the lymphocytic compartment, including spleen, thymus, peripheral blood lymphocytes and ovary.
Sequence similarities: Contains 9 C2H2-type zinc fingers.
Sequence caution: The sequence AAA98503.1 differs from that shown. Reason: Frameshift at positions 189, 212 and 1087.
The sequence AAH38985.1 differs from that shown. Reason: Miscellaneous discrepancy. Contaminating sequence. Potential poly-A sequence.
Molecular Weight~120 kDa observed; 121.87 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
