Background InformationTranscription factor 4 (UniProt: P15884; also known as TCF-4, Class B basic helix-loop-helix protein 19, bHLHb19, Immunoglobulin transcription factor 2, ITF-2, SL3-3 enhancer factor 2, SEF-2) is encoded by the TCF4 (also known as BHLHB19, ITF2, SEF2) gene (Gene ID: 6925) in human. TCF-4 is a member of the basic helix loop helix (bHLH) family of transcription factors that preferentially binds to either 5'-ACANNTGT-3' or 5'-CCANNTGG-3'. It is expressed in adult heart, brain, placenta, skeletal muscle and to a lesser extent in the lung. In developing embryonic tissues, its expression is mostly in the brain. TCF-4 plays an important role in several developmental processes, including the initiation of neuronal differentiation. The bHLH domain of TCF-4 is localized in amino acids 564-617 and contains a highly conserved group of positively charged amino acids that make up the basis region that interacts with DNA. The remaining part of bHLH forms a surface that favors its homo- and heterodimerization through the assembly of a stable four-helix bundle with a hydrophobic core. The bHLH domain permits TCF-4 to interact with several transcription factors. Its DNA-binding properties are shown to be regulated by Calcium binding proteins, such as calmodulin and S100 alpha and beta. Clone 6F12 is shown to react with both TCF-3 and TCF-4.
