Background InformationISEC1 protein, intestinal secretory cell protein 1 (UniProt: A0A077K8V5) is encoded by the Gm609 (also known as isec1) gene (Gene ID: 208166) in murine species. iSEC1 is a D200 homologue that is expressed mainly in the small intestine, but some expression is observed in the stomach, duodenum, jejunum, ileum, cecum, and colon. It is not detected in esophagus or other tissues. It can bind to CD200R, but not to other members of the CD200R family. Binding of iSEC1 to CD200R on intraepithelial lymphocytes (IELs) results in the suppression of cytokine production and cytolytic activity activated by IELs. Hence, iSEC1 is considered as a negative regulator of IELs function. iSEC1 is synthesized with a signal peptide (aa 1-23) that is cleaved off in the mature form. The mature form exists as a disulfide-linked homodimer. (Ref.: Kojima, T., et al. (2016). Sci. Rep. 6: 36457).
