Background InformationNeutrophil cytosol factor 1 (UniProt: P14598; also known as NCF-1, 47 kDa autosomal chronic granulomatous disease protein, 47 kDa neutrophil oxidase factor, NCF-47K, Neutrophil NADPH oxidase factor 1, Nox organizer 2, Nox-organizing protein 2, SH3 and PX domain-containing protein 1A, p47-phox) is encoded by the NCF1 (also known as NOXO2, SH3PXD1A) gene (Gene ID: 653361) in human. p47Phox is a peripheral membrane protein that is a component of an NADPH oxidase complex. In activated cells p47phox migrates to the membranes, where it associates with the membrane-bound component (flavocytochrome b558) to assemble the catalytically active oxidase. Its N-terminal contains one PX (phox homology) domain (aa 4-125), the middle amino acid sequence contains two SH3 domains (aa 156-215 and 226-285). Its COOH-terminal sequence has a basic charge and is rich in serine and arginine residues. The PX domain mediates interaction with phosphatidylinositol 3,4-bisphosphate and other anionic phospholipids. In resting state, its SH3 domains interact with the C-terminal region to keep p47phox in an auto-inhibited state and phosphorylation is shown to disrupt this auto-inhibited state. p47phox contains six serine phosphorylation sites and its phosphorylation on multiple serine plays a pivotal role in oxidase activation in intact cells. TNFalpha is reported to prime reactive oxygen species (ROS) production through phosphorylation of p47phox on serine 345. This site is phosphorylated by MEK1/2. S345A mutation is shown to block TNF and GM-CSF induced priming of ROS production. Mutations in NCF1 gene is known to cause Granulomatous disease that is characterized by the inability of neutrophils and phagocytes to kill ingested microbes. (Ref.: Boussetta, T., et al. (2009).
