Background InformationS6 (Ribosomal protein S6, RpS6, 40S ribosomal protein S6) is known to play a role in cell growth and proliferation. It lays downstream of both the PI3K/Akt/mTOR pathway as a substrate of p70 S6 kinase and the Ras/Erk pathway as a substrate of RSK. S6 is phosphorylated in response to stimulation by growth factors, tumor promoting agents, and mitogens. It is dephosphorylated at growth arrest. There are two major areas of S6 phosphorylation; Ser235/Ser236 that is known to be phosphorylated by both p70S6 Kinase as well as RSK as well as Ser240/244 which is thought to only be phosphorylated by p70S6K.
