Application NotesPeptide Inhibition Analysis:
10 μM of a Histone H3 peptide containing trimethyl-lysine 9 abolished detection of Histone H3 by anti-trimethyl Histone H3 (Lys9) in western blot analysis of HeLa acid extracts (Figure B, lane 4). Slight signal reduction was observed with 10 μM of a Histone H3 peptide containing dimethyl-lysine 9.

Peptide Dot Blot Analysis:
A 1:2000 dilution of a previous lot was incubated with serial dilutions of Histone H3 peptides containing various modifications spotted onto PVDF. Slight cross-reactivity with Histone H3 peptide containing dimethyl-lysine 9 was detected.

Beadlyte® Histone Peptide Assay:
1:2000-1:6000 dilutions of a previous lot were incubated with Histone H3 peptides containing various modifications conjugated to Luminex® microspheres. Slight cross-reactivity with Histone H3 peptides containing dimethyl-lysine 9 and trimethyl-lysine 27 was detected.

Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. Symmetric dimethylation at Arg-9 by PRMT5 is linked to gene repression (By similarity). & Methylation at Lys-5, Lys-37 and Lys-80 are linked to gene activation. Methylation at Lys-10 and Lys-28 are linked to gene repression.
