It is crucial that water used for these applications does not introduce any impurities that could interfere with analytical results. As modern analytical instruments become capable of detecting ever increasingly low concentrations of analytes, ensuring the lowest level of background interference has become even more important—and more difficult. To meet this challenge, laboratory water purification systems have evolved accordingly. Current systems such as the Milli-Q Integral use a sophisticated combination of purification technologies to reduce ions, organics and other potentially background-causing contaminants to extremely low levels (i.e., 18.2 MΩ.cm resistivity and <5 µg/mL of TOC). | Endotoxin-Free Water for Tissue Culture E. Riché, J. Bôle and S. Mabic Lab Manager Magazine (2010) May 2010 Show Abstract Water is a ubiquitous solvent used throughout cell isolation and culture. Therefore, water contamination may affect experimental outcomes. The use of ultrapure water is advisable for culturing of cells sensitive to environmental contaminants, such as endotoxins and bacteria. Cardiomycyte isolation experiments showed that using water contaminated with bacteria and endotoxins decreases cell viability, while using freshly produced ultrapure water dramatically increases it. Mabic Clinical Lab Products (2007) April, 22-23 2007 Show Abstract Sophisticated diagnostic equipment is designed to improve quality, increase throughput, and manage continued labor shortages. All diagnostic manufacturers can provide the end user with software specifications that simplify automatic sampling and predilution; ensure workflow efficiency with high-speed throughput and performance; provide accuracy in testing with precision optical systems; deliver real-time access and alerts for patient and QC packages; and offer mechanical alerts to any possible instrumentation failures. Additionally, troubleshooting instrumentation, even with the above solution, creates unique challenges for medical technologists. Instrumentation can alert, flag, and warn that something is problematic, but it takes an experienced medical technologist to determine the root cause. One parameter, even though utilized by each end user on any kind of diagnostic instrument, has not been considered: water. Used in a variety of assays, water is a major reagent in clinical chemistry and immunoassay testing. Analytical factors linked to water quality need to be controlled and optimized to reduce the number of test failures. The water quality delivered to the analyzer is as important as any other reagent. Control of bacteria and its by-products with Elix technology and Biopak filters provides the highest quality water to be used in assays sensitive to these contaminants. Control of the water quality eliminates frequent decontamination. A single purification unit can be connected to up to three point-of-use dispensers, which can be equipped with different purifiers to provide the necessary water quality for various applications. Biopak® Polisher is designed to be connected to the outlet of water purification systems such as the PODs of Milli-Q® systems. It is warranted for a period of up to three months. The ultrafilter is composed of polysulfone hollow fibers which structure has been optimized to efficiently reject targeted contaminants, while maintaining a high flow rate and minimizing the release of ionic and organic materials.
Pyrogen-free and bacteria-free water
Pyrogens are known to affect cell culture and biochemistry experiments in numerous ways. The most common pyrogens from bacteria are endotoxins (LPS -lipopolysaccharide). In ultrapure water, LPS sub-units aggregate to form high molecular weight structures that can be efficiently removed by ultrafiltration membranes. Experiments have validated that Biopak® Polisher can be used for at least 90 days* to purify ultrapure water to obtain endotoxin levels below 0.001 EU/mL.
Nuclease-free water
Molecular biologists systematically require nuclease-free reagents as nucleic acid molecules are at the heart of their experimental research. Biopak® ultrafiltration has been demonstrated as an effective and practical solution for these scientists. And, contrary to DEPC (diethylpyrocarbonate), ultrafiltration does not generate hazardous handling conditions, lengthy treatment time or contamination of the treated water by CO2 and ethanol. Challenge tests have validated Biopak® efficiency in removing RNases (< 1 pg/mL) and DNases (< 5 pg/mL) when connected to Milli-Q® water purification system. Each Biopak® Polisher is delivered with its Certificate of Quality.
