OverviewAll ChIPAb+ antibodies are individually validated for chromatin precipitation, every lot, every time. Each ChIPAb+ antibody set includes control primers (tested every lot by qPCR) to biologically validate your IP results in a locus-specific context. The qPCR protocol and primer sequences are provided, allowing researchers to validate ChIP protocols when using our antibody in their chromatin context. Each set also includes a negative control antibody to ensure specificity of the ChIP reaction.
The ChIPAb+ CHD1 validated antibody and primer set includes a CHD1 antibody, a Normal Rabbit IgG, and positive control primers which amplify 114 bp of human PSMC6 (proteasome 26S subunit 6) The CHD1 antibody and negative control IgG are supplied in a scalable "per ChIP" reaction size and can be used to functionally validate the precipitation of CHD1 associated chromatin.

Background InformationChromodomain-helicase-DNA-binding protein 1 (CHD1) is a multidomain chromatin-remodelling enzyme, which belongs to the SNF2/RAD54 helicase family. It contains an SWI/SNF-like helicase/ATPase domain, a DNA-binding domain, and two chromodomains. CHD1 is multifunctional protein, which has an important role in gene regulation. It interacts with various modified histones, such as H3K4Me2 and H3K4Me3, and maintains the open chromatin structure required for transcription. It also interacts with, or plays a role in the assembly of, several transcription-related complexes, including the histone acetylation complex SAGA, and the FACT and PAF complexes. Previous studies have suggested that CHD1 maintains the pluripotent state of embryonic stem cells, and is required for the reprogramming of somatic cells to the pluripotent state. CHD1 may play a role in the development of prostate cancer.

Sonicated chromatin prepared from K562 cells (~4e6 cell equivalents per IP) were subjected to chromatin immunoprecipitation using 0.7 µg of either Normal Rabbit IgG (Part No. PP64B), or 0.7 µg Anti-CHD1 (Part No. CS216151) and the Magna ChIP™ HiSens (Cat. # 17-10460). Successful immunoprecipitation of CHD1 associated DNA fragments was verified by qPCR using ChIP Primers, PSMC6 (Part No. CS216152) as a positive locus, and ChIP Primers, SNRPN (Part No. CS207347) as a negative locus.

Chromatin Immunoprecipitation:
