Recommended reaction conditions
Protocol for Coating Tissue Culture Plates With Fibronectin Note: this protocol is meant to serve as a guideline. Optimization may be required for individual cell types.
Note: This protocol is based on the use of 21 cm2 dishes (Corning 25010, Costar 3060, Falcon 3002, or an equivalent 60 mm dish) and 1 mg fibronectin; 1 mg fibronectin is sufficient for coating 10 dishes at 5 µg/cm2.
1. Thaw the fibronectin by placing the vial in a 37°C water bath and leaving it undisturbed until completely thawed. DO NOT DISTURB THE VIAL AT ANY TIME DURING THE THAWING PROCESS! If the vial is disturbed or removed prior to complete thawing, the fibronectin will form a gel and be unusable. DO NOT VORTEX. DO NOT SHAKE AFTER THAWING. Mix very gently after thawing. 2. After thawing, bring the solution to a final volume of 20 ml with sterile, serum-free culture medium that has been pre-warmed to 37°C. This yields a fibronectin solution of 50 µg/ml. 3. Add 2 ml of this solution to each of 10 tissue culture dishes and swirl gently to completely coat the growth surface. 4. Allow the dishes to remain at room temperature for ~45 min to permit binding of the fibronectin to the surface of the dish. 5. Tilt each dish and remove the fibronectin using a sterile pipet. Do not permit the pipet tip to touch or disturb the growth surface. 6. Add the cell suspension in medium directly to the fibronectin-coated dish.
