OverviewEMD Millipore’s MILLIPLEX® MAP Human IL-18 Singleplex Kit is validated for detection in serum, plasma or tissue/cell supernatant samples, and for use as follows:
• On its own as a single protein quantification assay, Cat. No. HIL18MAG-66K (the standard is calibrated to the WHO standard)
• Combined with MILLIPLEX® MAP Human Cytokine/Chemokine Panel 1, Cat. No. HCYTOMAG-60K (or the corresponding premix or bulk format kits)
• Combined with MILLIPLEX® MAP Human Cytokine/Chemokine Panel 2, Cat. No. HCYP2MAG-62K (with the exception of the CTACK/CCL27 assay, due to bead region overlap)
• Combined with MILLIPLEX® MAP Human Th17 Panel, Cat. No. HTH17MAG-14K (or the corresponding premix or bulk format kits)
Background InformationInterleukin-18 (IL-18, also known as interferon-gamma inducing factor) is a member of the IL-1 cytokine superfamily and acts as a pleiotropic cytokine that modulates both innate and acquired immune responses. Disrupted regulation of IL-18 bioactivity is linked to multiple types of diseases including chronic inflammatory diseases, autoimmune diseases, a variety of cancers, and infectious diseases. It also has been implicated to play a role in emphysema, myocardial function, cardiovascular disease, metabolic syndrome, acute kidney injury and neurodegeneration. Our MILLIPLEX® MAP Human IL-18 Singleplex Magnetic Bead Kit is a single analyte kit to be used for the quantification of IL-18 in serum, plasma, and tissue/cell supernatant samples. The Human IL-18 Singleplex standard is calibrated to the WHO standard.

Application Notes• A maximum of 25 μL per well neat serum or plasma can be used. Tissue culture or other media may be run neat or appropriately diluted in control medium.
• As single analyte assay, this assay may be run as a 2 hour or overnight incubation assay. If combining with one of the other indicated panels, please follow the protocol for that particular panel.
• When running this assay as a single analyte assay, follow the protocol directions and use the reagents that come in the HIL18MAG-66K kit.
• When combining the IL-18 analyte with one of the larger panels, use the reagents provided in the other kit. Carefully reference both protocols to ensure proper procedures are followed, particularly for the following sections:
o “PREPARATION OF REAGENTS FOR IMMUNOASSAY: HUMAN IL-18 WITH OTHER KITS”
- Note: Use the serum matrix (if needed) provided with the larger panel
o Follow the “IMMUNOASSAY PROCEDURE” for the larger panel
• If comparing single analyte IL-18 sample values with IL-18 combined with one of the possiible kitss, you must run a curve to determine sample correlation and slope. Differences in sample values are expected due to differences in serum matrix, but should highly correlate.
