Background InformationCollagen alpha-1(XVII) chain (UniProt: Q9UMD9; also known as 180 kDa bullous pemphigoid antigen 2, Bullous pemphigoid antigen 2, COL17A1) is encoded by the COL17A1 (also known as BP180, BPAG2) gene (Gene ID: 1308) in human. COL171A1 is a single pass type II membrane protein that is expressed in skin, cornea, bladder, thymus, and esophagus. It has a cytoplasmic domain (aa 1-467), a transmembrane domain (aa 468-488), and a long extracellular domain (aa 489-1497). It is shown to exclusively localized to anchoring filaments. Following its synthesis, it is cleaved into two chains: a 120 kDa linear IgA dermatosis antigen (LAD-1) and a 97-kDa linear IgA bullous dermatosis antigen (97-LAD). Both these chain of COL17A1 represent major antigenic targets of autoantibodies in subjects with Linear IgA disease (LAD), a subepidermal blistering disorder. Clone 123 is reported to recognize the 120 kDa protein from conditioned cell culture medium and a 97 kDa protein in human skin extracts. (Ref.: Marinkovich, MP., et al. (1996). J. Invest. Dermatol. 106(4); 734-738).
