      Background InformationFractalkine (UniProt: P78423; also known as C-X3-C motif chemokine 1, CX3C membrane-anchored chemokine, Neurotactin, Small-inducible cytokine D1, CXCL1) is encoded by the CXCL1 (also known as FKN, NTT, SCYD1, A-152E5.2) gene (Gene ID: 6376) in human. CX3CL1 is a monomeric, single-pass type I membrane protein that acts as a ligand for CX3CR1 and integrins. It is expressed in multiple tissues and the seminal plasma, endometrial fluid, and follicular fluid. Its highest levels have been reported in the brain and heart. It is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 25-341), a transmembrane domain (aa 342-362), and a cytoplasmic domain (aa 363-397). It can activate integrins in both a CX3CR1-dependent and independent manners. In the presence of CX3CR1, it activates integrins by binding to their classical ligand-binding site (site 1) and in the absence of CX3CR1, it binds to a second site (site 2) in integrins. Binding to site 2 enhances the binding of other integrin ligands to site 1. CX3CL1 can exist in bot soluble and membrane-bound form. The membrane-bound form is proteolytically cleaved by ADAM10 and ADAM17 to generate the soluble form. The soluble form is shown to be chemoattractive for T-cells and monocytes and the membrane-bound form promotes adhesion of those leukocytes to endothelial cells. The membrane-bound form can be induced on primary endothelial cells by inflammatory cytokines such as TNF- , interferon- (IFN- ), and interleukin-1. (Ref.: Jones, BA., et al, (2010).
