      Background InformationDNA ligase 3 (EC 6.5.1.1; UniProt P49916; also known as DNA ligase III, ATP-dependent polydeoxyribonucleotide synthase [ATP] 3) is encoded by the LIG3 gene (Gene ID 3980) in human. Three genes (LIG1, LIG3, and LIG4) in human genome encode ATP-dependent DNA ligases required for repairing DNA single- and double-strand breaks (SSB and DSB) and completing repair events initiated by the excision of mismatched and damaged bases. Among the human LIG genes, the LIG3 is unique in that it encodes multiple isforms with different N- and C-termini via alternative translation initiation and splicings. The "breast cancer susceptibility protein 1 C-terminal" (BRCT) domain present at the C-terminal end of the canonical isoform 1 (a.a. 933-1009) and isoform 3 is replaced by a short positively charged sequence that acts as a nuclear localization signal in spliced isoform 2 and isoform 4 (also known as the beta isoform). In addition, the N-terminal end of isoform 3 (also known as the alpha isoform) and isoform 4 do not contain the 87 a.a. mitochondrial targeting sequence present in isoform 1 and isoform 2. The LIG3 is the only gene encodes mitochondria DNA ligase (i.e. isoforms 1 & 2), inactivation of the LIG3 gene results in loss of mitochondrial function and cell death. Isoforms 1 and 3 interact with the scaffold protein XRCC1 via their BRCT domain, the XRCC1 in turn recruits a large number of proteins involved in base excision and single-strand break repair.
