      Background InformationProblems in the CNS dopaminergic system are thought to be the root cause of some neuropsychiatric disorders. Dopamine D2 receptor is one of the five types (D1 to D5) of receptors for dopamine. The activity of this receptor is mediated by G proteins which inhibit adenylyl cyclase. Dopamine D2 receptor signaling using Nurr1 and ERK may play an important role in mesencephalic dopaminergic neuronal development. Faulty D2 receptors have been common in cases of ROS-dependent hypertension. In addition, defects in DRD2 are associted with alcohol-responsive dystonia, varying susceptability to familial alcoholism as well as a contributing factor to the suicide risk in alcoholics.

      DISEASE: It has been suggested that DRD2 is involved in psychiatric disorders; especially in schizophrenia. & Defects in DRD2 are associated with myoclonus dystonia (MD) [MIM:159900]; also known as myoclonic dystonia, alcohol- responsive dystonia and dystonia-11 (DYT11). MD is a movement disorder characterized by involuntary lightning jerks and dystonic movements and postures alleviated by alcohol. Inheritance is autosomal dominant. The age of onset, pattern of body involvement, presence of myoclonus and response to alcohol are all variable.
