      Background InformationGasdermin-D (UniProt: P57764; also known as Gasdermin domain-containing protein 1) is encoded by the GSDMD (also known as DFNA5L, GSDMDC1, FKSG10) gene (Gene ID: 79792) in human. Gasdermin-D is a member of the gasdermin family of proteins that promotes inflammatory cell death (pyroptosis) in response to microbial infection and danger signals. Its expression is observed in the suprabasal cells of esophagus, as well as in the isthmus/neck, pit, and gland of the stomach. Gasdermin-2 consists of a 31 kDa N-terminal and a 22 kDa C-terminal domains that are separated by a linker region. Under normal conditions, the full-length Gasdermin-D is inactive as the linker loop between the N-terminal and C-terminal domains stabilizes the overall conformation of the full-length protein and allows the C-terminal domain to fold back on and auto-inhibit the N-terminal Gasdermin-D (Gasdermin-N). Upon interdomain cleavage by the action of caspase-1 and 4 at Asp275, the autoinhibition is relieved and Gasdermin-N cytotoxicity is triggered. Gasdermin-N is reported to form membrane pore that lead to cytokine release and pyroptosis. It also displays bactericidal activity. Gasdermin-N released from pyroptotic cells into the extracellular milieu can bind to and kills both Gram-negative and Gram-positive bacteria, without harming neighboring mammalian cells.
