Background InformationInterferon-induced GTP-binding protein Mx2 (UniProt P20592; also known as Interferon-regulated resistance GTP-binding protein MxB, Myxovirus resistance protein 2, p78-related protein, Second interferon-induced protein p78) is encoded by the MX2 (also known as MXB) gene (Gene ID 4600) in human. The interferon-inducible myxovirus resistance (Mx) proteins belong to the family of large GTPases and are highly homologous with dynamins within their GTP-binding domain. MxA/Mx1 is cytosolic, while two MxB/Mx forms exist, a 78 kDa nuclear form and a 76 kDa cytosolic form lacking the N-terminal nuclear localization signal (NLS). Human MxA protein mediates resistance to influenza and several other viruses, while human MxB is also reported to inhibit HIV-1 infection by reducing the level of integrated viral DNA. In addition, HIV-1 with Ala88 mutation in the capsid protein (CA) is found to escape MxB restriction, as a result of impaired CA interaction with the host peptidylprolyl isomerase cyclophilin A (CypA). Likewise, CypA deletion results in the loss of MxB inhibition of HIV-1.

      Applications
ApplicationThis Anti-MxB Antibody is validated for use in Western Blotting for the detection of MxB.
      Key Applications
      • Western Blotting
Application NotesWestern Blotting Analysis: A representative lot detected IFNalpha-2b-induced MxB/Mx2 expression in U87-MG human glioblastoma cells and SupT1 human T lymphoblast cells (Liu, Z., et al. (2013). Cell Host Microbe. 14(4):398-410).
Western Blotting Analysis: A representative lot detected doxycycline-induced MxB/Mx2 expression using virally transformed human T lymphoblast SupT1 cells that stably expresses the reverse tetracycline-controlled transactivator (rtTA) protein and carries the MxB gene under the tetracycline-responsive promoter element/TRE (Liu, Z., et al. (2013). Cell Host Microbe. 14(4):398-410).
