      Background InformationThe thymus is considered as the primary lymphoid organ that is responsible for the generation and maturation of T cells. Thymic epithelial cells (TECs) account for the majority of thymic stromal components. These cells can be divided into cortical and medullary thymic epithelial cells based on their localization within the thymus. TECs have dual germ layer origins, with cortical cells arising from ectoderm and medullary cells arising from the endoderm. In an embryonic thymus, cortical cells are the dominant population, while medullary cells account for most of the TECs in the adult thymus. Cortical thymic epithelial cells are essential for the positive selection of T cells, whereas the medullary thymic epithelial cells (mTECS) play a role in inducing negative selection of highly reactive T cells that are required for establishing central self-tolerance. mTECs participate in self-tolerance by eliminating self-reactive T cells. Cortical cells that support positive selection also support post-selection maturation of single-positive thymocytes. They provide cytokines, such as IL-7 and SCF, to promote early thymic progenitors (ETP) cells and Delta-like ligand 4 (DLL4)-mediated Notch signaling to induce the differentiation of ETPs towards the T lineage. DLL4 is reported to be expressed at high levels in cortical cells, but not in medullary cells. Clone ER-TR4 reacts with epithelial-reticular cells in the cortex of murine thymus.
