      Background InformationPlasmalemmal neurotransmitter transporters sequester synaptic and peri synaptic transmitter into presynaptic elements. The Dopamine Transporter (DAT), a 12 transmembrane domain transporter with the N and C terminal regions located within the cytoplasm is responsible for the reaccumulation of dopamine after it has been released. Levels of DAT protein expression are altered by chronic drug administration. The activity of the DAT reuptake carrier is sodium dependent, and it is suspected to play a role in such neurologic and psychiatric disorders as Parkinson's disease, Tourette's disease, schizophrenia, and addiction. DAT is also the target of several “DAT-blockers” including amphetamines and cocaine. These chemicals inhibit the action of DAT and, to a lesser extent, the other monoamine transporters, but their effects are mediated by separate mechanisms.
