      Background InformationEF-hand calcium-binding domain-containing protein 9 (UniProt: Q9DAM2; also known as EFCAB9) is encoded by the Efcab9 gene (Gene ID: 69306) in murine species. EFCAB9 is an evolutionarily conserved, testis-specific protein that is expressed after meiosis. It serves as a bifunctional cytoplasmic machine that modulates the channel activity and the domain organization of CatSper. It is a pH-dependent calcium sensor that is required for activation of CatSper channel required for sperm cell hyperactivation. This hyperactivation is essential for sperm motility and fertilization. It associates with the CatSper complex via direct interaction with subunit CATSPERz, and senses intracellular Calcium levels. However, it dissociates at elevated pH. It is shown that EFCAB9-CatSperz protein pairs interact with the channel pore as a gatekeeper before capacitation, but it activates the channel in response to capacitation-associated intracellular pH and changes in calcium levels. ECFCAB9 is reported to contain three conserved EF-hand Calcium binding domains. Mice deficient in EFCAB9 display sperm motility defects and compromised fertility.
