      Background InformationGuanine nucleotide-binding protein G(t) subunit alpha-1 (UniProt P11488; also known as Transducin alpha-1 chain) is encoded by the GNAT1 (also known as GNATR, CSNB1G, CSNBAD3) gene (Gene ID 2779) in human. Heterotrimeric G proteins are activated by G protein-coupled receptors (GPCRs) and are composed of alpha (α), beta (β), and gamma (γ) subunits. There exists also monomeric Ras superfamily of small GTPases that are commonly referred to as small G-proteins. G-proteins mediate signal transduction initiated by GPCRs. Ligand binding activates GPCRs by inducing a conformational change. The activated GPCRs catalyze the GDP-to-GTP exchange of the Gα subunit, causing a dissociation of the Gβγ dimer from the now GTP-bound Gα subunit. The Gα subunit gradually hydrolyzes the bound GTP to GDP and eventually re-associates with Gβγ and re-starts the cycle. There exist at least 20 different Gα subunits, which are separated into four main groups, Gi, Gs, Gq, and G12/13. Eight different genes code for Gi alpha subunits, including GNAT1 and GNAT2 that code for Gt in the rod (rTα by GNAT1) and cone (cTα by GNAT2) cells of the retina, where they function as the alpha subunit of the trimeric G protein transducin. The α-subunit binds GTP and activates cyclic GMP phosphodiesterase (PDE), whereas the β and γ subunits mediate interaction with rhodopsin. Mutations in RHO, PDE6B, and GNAT1 genes are associated with autosomal dominant congenital stationary night blindness (CSNB), and patients with mutations in these genes exhibit the Riggs- or Schubert-Bornschein-type electronegative electroretinogram (ERG).
