General description

FUNCTION: SwissProt: O75311 # The glycine receptor is a neurotransmitter-gated ion channel. Binding of glycine to its receptor increases the chloride conductance and thus produces hyperpolarization (inhibition of neuronal firing).
SIZE: 464 amino acids; 53800 Da
SUBUNIT: Pentamer composed of alpha and beta subunits (By similarity).
SUBCELLULAR LOCATION: Cell junction, synapse, postsynaptic cell membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Widely distributed throughout the central nervous system.
DOMAIN: SwissProt: O75311 The N-terminal domain carries structural determinants essential for agonist and antagonist binding. TM2 is thought to form the innert wall of the chloride channel. The cytoplasmic loop is an important determinant of channel inactivation kinetics.
SIMILARITY: Belongs to the ligand-gated ionic channel (TC 1.A.9) family.
MISCELLANEOUS: The alpha subunit binds strychnine.
A purified peptide corresponding to amino acid residues 355-368 of rat GlyRa3 (Accession P24524).

Specificity

Recognizes Glycine receptor alpha3 (GlyRalpha3) protein. The epitope is not similar to any other known proteins.

Application

Western blot: 1:200 on rat brain membranes.

Immunohistochemistry on rat brain floating sections and mouse brain floating sections (trypsin pretreatment is recommended).

Dilutions should be made using a carrier protein such as BSA (1-3%)

Optimal working dilutions must be determined by the end user.

Physical form

Affinity purified immunoglobulin. Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA, 5% sucrose as a stabilizer and 0.05% sodium azide as a preservative. Reconstitute with 200 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).
Affinity purified immunoglobulin. Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA and 0.05% sodium azide as a preservative. Reconstitute with 50 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).

Storage and Stability

Maintain lyophilized material at -20°C for up to 12 months after date of receipt. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

CONTROL ANTIGEN: Included free of charge with the antibody is 20 μg of control antigen (lyophilized powder). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For negative control, preincubate 1 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
