      DescriptionAnti-Histone H3Kme3Q5Ser
      Alternate Names
      • H3KmeQ5Ser
Background InformationHistone H3.1 UniProt: Q16695; also known as H3/t, H3t, H3/g) is encoded by the HIST3H3 (also known as H3FT) gene (Gene ID: 8290) in human. Histone H3 has two main variants, H3.1 and H3.3, which show different genomic localization patterns in animals. Histone H 3.1 is a core component of nucleosome that is present only in mammals. It is expressed during S phase, then its expression decreases significantly as cell division slows down during the process of differentiation. Histone H 3.1 expression is shown to be replication dependent. It serves as the canonical histone, which is incorporated during DNA replication. Histone 3.1 can also undergo serotonylation of glutamine at position 5 (Q5ser) in organisms that produce serotonin. This has been demonstrated that tissue transglutaminase 2 can serotonylate histone H3K4me3 marked nucleosomes, which results in the formation of H3K4me3Q5ser. The presence of Q5ser does not affect the ability of MLL1, an H3K4 methyltransferase, to modify nucleosomes in vitro. In the brain the H3K4me3Q5ser signal is not segregated only to regions in which serotonin is produced, but is shown to be broadly distributed. H3K4me3Q5ser expression has also been reported in human pluripotent stem cell-derived serotonergic neurons and with differentiation the levels of H3K4me3Q5ser increase significantly.

      Applications
ApplicationAnti-Histone H3Kme3Q5Ser, Cat. No. ABE2580, is a highly specific rabbit polyclonal antibody that targets Histone H3 trimethylated on Lysine 4 and serotonylated on glutamine 5 and has been tested for use in ChIP-seq, Dot Blot, and Western Blotting.
      Key Applications
      • ChIP-seq
      • Dot Blot
      • Western Blotting
Application NotesChIP-seq Analysis: A representaive lot detected Histone H3Kme3Q5Ser in ChIP-seq applications (Farrelly, L.A., et. al. (2019). Nature. 567(7749):535-539).

Western Blotting Analysis: A 1:500 dilution from a representative lot detected Histone H3.1t in whole cell lysates from mouse brain across multiple regions (hippocampus, prefrontal cortex, ventral tegmental area, dorsal raphe nucleus, nucleus accumbens, caudate putamen, cerebellum). (Courtesy of Dr. Ian Maze, Icahn School of Medicine at Mount Sinai, New York).

Western Blotting Analysis: A 1:500 dilution from a representative lot detected Histone H3Kme3Q5Ser in whole cell lysate from Undifferentoated & differentiated RN46a-B14 cells.

Western Blotting Analysis: A representaive lot detected Histone H3Kme3Q5Ser in Western Blotting applications (Farrelly, L.A., et. al. (2019). Nature. 567(7749):535-539).

ChIP-seq Analysis: A representative lot detected Histone H3Kme3Q5Ser in H3K4me3Q5ser at Fos (brain expressed) vs. Hpx (liver specific gene) loci showing nice enrichment in brain over DNA input (mouse dorsal raphe nucleus, adult). (Courtesy of Dr. Ian Maze, Icahn School of Medicine at Mount Sinai, New York).

Dot Blot Analysis: Unmodified and various modified Histone peptides were probed with H3Kme3Q5Ser (Courtesy of Dr. Ian Maze, Icahn School of Medicine at Mount Sinai, New York).
