StAR-related lipid transfer protein 3 (UniProt: Q14849; also known as Metastatic lymph node gene 64 protein, MLN 64, Protein CAB1, START domain-containing protein 3, StARD3) is encoded by the STARD3 (also known as CAB1, MLN64) gene (Gene ID: 10948) in human. StARD3 is a multi-pass, homodimeric membrane protein that localizes to contact sites between the endoplasmic reticulum and late endosomes. It is a sterol-binding protein that mediates cholesterol transport from the endoplasmic reticulum to endosomes and creates contact site between the endoplasmic reticulum and late endosomes. It acts as a lipid transfer protein that redirects sterol to the endosome at the expense of the cell membrane and favors membrane formation inside endosomes. It may also mediate cholesterol transport between other membranes, such as the mitochondrial membrane or cell membrane. StARD3 contains a START domain (aa 230-443) that mediates lipid-transfer between membranes and has a hydrophobic cavity that can accommodate one lipid molecule, thereby serving as a ′hydrophobic bridge′ across the aqueous gap between donor and acceptor organelle membranes. It also has a MENTAL domain (aa 46-217) that anchors the protein in endosome membranes and exposes the START domain in the cytosol. StARD3 is shown to interact with VAMP-associated proteins A and B (VAPA and VAPB) via its FFAT (two phenylalanines in an acidic tract (FFAT) motif (aa 207-212). (Ref.: Di Mattia, T., et al. (2018). Evaluated by Immunocytochemistry in SK-BR-3 cell line.

Immunocytochemistry Analysis: A 1:250 dilution of this antibody detected MLN64/STARD3 in SK-BR-3 cells.
