      Background InformationType 2 protein serine/threonine phosphatase (PP2A) is a trimer composed of a catalytic C subunit and two regulatory (A and B) subunits; many isoforms for the A and B subunits have been found. PP2A has a high specific activity against the α subunit of phosphorylase kinase and is insensitive to Inhibitor-2. PP2A is distinguishable from PP2B/Calcineurin and PP2C by its partial activity in the absence of divalent cations, and by its high sensitivity to inhibition by okadaic acid. It is also thought to be the principal downmodulator of serine/threonine kinase cascades. PP2B requires Ca2+/calmodulin for activity, and is an activator of NFAT transcription factors. PP2C requires Mg2+, but is insensitive to okadaic acid, microcystin LR, or the calmodulin inhibitor trifluoperazine.

      Application NotesImmunoprecipitation:
      4 µg of a previous lot immunoprecipitated PP2A from 500 µg of A431 RIPA lysate. Note: This antibody has been reported not to block phosphatase activity.

      Immunocytochemistry:
      5-10 µg/mL of a previous lot of this antibody showed positive immunostaining for PP2A in A431 cells fixed for 10 minutes with 4% paraformaldehyde in PBS.

      UniProt SummaryFUNCTION: SwissProt: P67775 # PP2A can modulate the activity of phosphorylase B kinase casein kinase 2, mitogen-stimulated S6 kinase, and MAP-2 kinase. Can dephosphorylate SV40 large T antigen and p53. Dephosphorylates SV40 large T antigen, preferentially on serine residues 120, 123, 677, and perhaps 679. The C subunit was most active, followed by the AC form, which was more active than the ABC form, and activity of all three forms was strongly stimulated by manganese, and to a lesser extent by magnesium. Dephosphorylation by the AC form, but not C or ABC form is inhibited by small T antigen.

Proteins that associate with the core dimer include three families of regulatory subunits B (the R2/B/PR55/B55, R3/B''/PR72/PR130/PR59 and R5/B'/B56 families), the 48 kDa variable regulatory subunit, viral proteins, and cell signaling molecules.
      SUBCELLULAR LOCATION: Cytoplasm.
      PTM: Reversibly methyl esterified on Leu-309. Carboxyl methylation may play a role in holoenzyme assembly. It varies during the cell cycle. & Phosphorylation of either threonine (by autophosphorylation- activated protein kinase) or tyrosine results in inactivation of the phosphatase. Auto-dephosphorylation has been suggested as a mechanism for reactivation.
