Background InformationHistone H4 (UniProt: P62805 ) is encoded by the HIST14A (also known as H4/A, H4FA, HIST1H4B, H4/I, H4FI, HIST1H4C, H4/G, H4FG, HIST1H4D, H4/B, H4FB, HIST1H4E, H4/J, H4FJ, HIST1H4F, H4/C, H4FC, HIST1H4H, H4/H, H4FH, HIST1H4I, H4/M, H4FM, HIST1H4J, H4/E, H4FE, HIST1H4K, H4/D, H4FD, HIST1H4L, H4/K, H4FK, HIST2H4A, H4/N, H4F2, H4FN, HIST2H4, HIST2H4B, H4/O, H4FO, HIST4H4) gene (Gene ID: 121504, 554313, 8294, 8359, 8360, 8361, 8362, 8363, 8364, 8365, 8366, 8367, 8368, 8370) in human. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Histone H4 is one of the main histone proteins that contribute to the chromatin structure in both prokaryotic and eukaryotic cells. Histone H4 can undergo several different types of epigenetic modifications that influence cellular processes, including methylation, acetylation, and phosphorylation. Acetylation at Lys6, Lys9, Lys13 and Lys17 is shown to occur in coding regions of the genome but not in heterochromatin. Monomethylation and asymmetric dimethylation at Arg3 is reported to favor acetylation at Lys9 and Lys13. Symmetrical dimethylarginine of histone H4 (H4R3me2s) is reported to serve as a bridge between DNA damage and subsequent repair. Intracellular H4R3me2s is shown to increase after treatment with the DNA oxidant, such as hydrogen peroxide. Histone H4 can be phosphorylated by Tie2 on Tyr51, which is shown to preferentially occur in the presence of core histones. The phosphorylated H4 (H4Y51) is recognized by the proto-oncogene ABL1. Histone H4 is also phosphorylated on Ser48 by PAK2, which is shown to increase the association of H3.3-H4 with the histone chaperone HIRA, thus promoting nucleosome assembly of H3.3-H4 and inhibiting nucleosome assembly of H3.1-H4. Chromosomal aberrations involving histone H4 causes of B-cell non-Hodgkin lymphomas. (Ref.: Ma, Z., et al. (2020). Redox Biol. 37; 101653; Hossain, MB., et al. (2016). Sci. Adv. 2; e1501290).

      Entrez Gene Number
Entrez Gene SummaryHistones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. This structure consists of approximately 146 bp of DNA wrapped around a nucleosome, an octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in a histone cluster on chromosome 1. This gene is one of four histone genes in the cluster that are duplicated; this record represents the telomeric copy. [provided by RefSeq].

      UniProt Number
UniProt SummaryFUNCTION: Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.

SUBUNIT STRUCTURE: The nucleosome is a histone octamer containing two molecules each of H2A, H2B, H3 and H4 assembled in one H3-H4 heterotetramer and two H2A-H2B heterodimers. The octamer wraps approximately 147 bp of DNA.

      SUBCELLULAR LOCATION: Nucleus. Chromosome.

PTM: Acetylation at Lys-6 (H4K5ac), Lys-9 (H4K8ac), Lys-13 (H4K12ac) and Lys-17 (H4K16ac) occurs in coding regions of the genome but not in heterochromatin.

Citrullination at Arg-4 (H4R3ci) by PADI4 impairs methylation.

      Monomethylation at Arg-4 (H4R3me1) by PRMT1 favors acetylation at Lys-9 (H4K8ac) and Lys-13 (H4K12ac). Demethylation is performed by JMJD6.

Symmetric dimethylation on Arg-4 (H4R3me2a) by the PRDM1/PRMT5 complex may play a crucial role in the germ-cell lineage.

Monomethylated, dimethylated or trimethylated at Lys-21 (H4K20me1, H4K20me2, H4K20me3). Monomethylation is performed by SET8. Trimethylation is performed by SUV420H1 and SUV420H2 and induces gene silencing.

Ubiquitinated by the CUL4-DDB-RBX1 complex in response to ultraviolet irradiation. This may weaken the interaction between histones and DNA and facilitate DNA accessibility to repair proteins. Monoubiquitinated at Lys-92 of histone H4 (H4K91ub1) in response to DNA damage. The exact role of H4K91ub1 in DNA damage response is still unclear but it may function as a licensing signal for additional histone H4 post-translational modifications such as H4 Lys-21 methylation (H4K20me).

Sumoylated, which is associated with transcriptional repression.

SEQUENCE SIMILARITIES: Belongs to the histone H4 family.

SEQUENCE CAUTION: The sequence AAI28106.1 differs from that shown. Reason: Frameshift at position 3.
Molecular Weight~13 kDa observed; 11.37 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
