      Background InformationCyclin-dependent kinase inhibitor 1 (UniProt: P38936; also known as CDK-interacting protein 1, Melanoma differentiation-associated protein 6, MDA-6, p21) is encoded by the CDKN1A (also known as CAP20, CDKN1, CIP1, MDA6, PIC1, SDI1, WAF1) gene in human. p21(WAF1) is a key mediator of p53-dependent cell cycle arrest and acts as a tumor suppressor. It is expressed in all adult tissues, however, its levels in the brain tissue are about 5-fold lower. It binds to and inhibits cyclin-dependent kinases, preventing phosphorylation of critical cyclin-dependent kinase substrates and blocking cell cycle progression. It binds tightly to the G1 and S phase kinases, cyclin E/Cdk2, cyclin D/Cdk4, and cyclin A/Cdk2, and effectively inhibits their activity. However, its effect on G2/M kinases is minimal. It binds the cyclin subunit through a conserved Cy1 motif in the N-terminal half and through a weaker and Cy2 motif in the C-terminal half. Its cyclin-binding region is localized to amino acids 17-24. Amino acids 53-58 are reported to be essential for its binding to CDKs. p21 inhibits proliferating cell nuclear antigen (PCNA) and binds to and blocks the inactivation of retinoblastoma protein (Rb), which is essential for cell cycle progression. Phosphorylation of p21 at Thr-145 by Akt or at Ser-146 by PKC is shown to impair its binding to PCNA. Its phosphorylation of Thr-145 by PIM2 is reported to enhance its stability and inhibits cell proliferation. In addition, phosphorylation of Thr-145 by PIM1 results in its relocation to the cytoplasm, which also adds to its stability. (Ref.: Abbas, T., and Dutta, A (2009). Nat. Rev. The encoded protein binds to and inhibits the activity of cyclin-CDK2 or -CDK4 complexes, and thus functions as a regulator of cell cycle progression at G1. The expression of this gene is tightly controlled by the tumor suppressor protein p53, through which this protein mediates the p53-dependent cell cycle G1 phase arrest in response to a variety of stress stimuli. This protein can interact with proliferating cell nuclear antigen (PCNA), a DNA polymerase accessory factor, and plays a regulatory role in S phase DNA replication and DNA damage repair. This protein was reported to be specifically cleaved by CASP3-like caspases, which thus leads to a dramatic activation of CDK2, and may be instrumental in the execution of apoptosis following caspase activation. Two alternatively spliced variants, which encode an identical protein, have been reported.
