      Background InformationMitogen-activated protein kinase 14 (UniProt: Q16539; also known as MAP kinase 14, MAPK 14, Cytokine suppressive anti-inflammatory drug-binding protein, CSAID-binding protein, CSBP, MAP kinase MXI2, MAX-interacting protein 2, Mitogen-activated protein kinase p38 alpha, MAP kinase p38 alpha, p38 alpha, Stress-activated protein kinase 2a, SAPK2a) is encoded by the MAPK14 (also known as CSBP, CSBP1, CSBP2, CSPB1, MXI2, SAPK2A) gene (Gene ID: 1432) in human. p38alpha is a serine/threonine kinase that acts as an essential component of the MAP kinase signal transduction pathway. It is one of the four p38 MAPKs that play an important role in the cascades of cellular responses evoked by proinflammatory cytokines or physical stress leading to direct activation of transcription factors. p38 MAPKs phosphorylate a broad range of proteins and it has been estimated that they may have approximately 200 to 300 substrates each. p38alpha is activated by cell stresses such as DNA damage, heat shock, osmotic shock, and pro-inflammatory stimuli. Its activation occurs through dual phosphorylation of Thr-180 and Tyr-182 by either of two dual specificity kinases, MAP2K3/MKK3 or MAP2K6/MKK6, and potentially also MAP2K4/MKK4. When phosphorylated on both Thr-180 and Tyr-182 it is shown to be about 10-20-fold more active than phosphorylation only on Thr-180. Phosphorylation of p38alpha on Tyr-182 alone keeps it in inactive state. The protein kinase domain of p38alpha is localized to amino acids 24-308.
