      DescriptionMillicell® µ-Angiogenesis Inhibition Assay Kit
      OverviewOverview

Studying how compounds affect angiogenesis, either to promote or inhibit new capillary tube formation can lead to therapies affecting wound healing, tissue regeneration, cardiovascular disease, stroke, tumor progression, and more. The Millicell μ-Angiogenesis activation and inhibition kits provide a powerful, quantitative platform for real-time monitoring of changes in tubule formation with unprecedented optical resolution.
      Benefits

      • Continuous, live monitoring of cells as tubules form
      • Even, flat slide surface prevents any liquid meniscus from forming, eliminating out-of-focus areas
      • Full well visualization at low magnification
      • Optimized assay conditions in a miniaturized format
      • Compatible with multi-channel pipettes and fixation reagents
      • Less reagents and cells needed, reducing costs and waste by up to 80%
      • All-in-one slide used to grow, assay, and stain cells

Background InformationAngiogenesis, the formation of new blood vessels from a pre-existing vascular network, occurs normally in development and is critical for a majority of the vessel formation that occurs during embryogenesis, tissue generation, and wound healing. However, abnormal blood vessel growth, either excessive or insufficient, can be the underlying cause of many deadly and debilitating diseases including cancer, cardiovascular disease, stroke, and diabetic and age-related blindness. Identification of specific compounds that promote or inhibit angiogenesis may provide promising treatments for these diseases.

Tubular formation is a multi-step process involving cell adhesion, migration, differentiation, and growth. Millipore’s Millicell® µ-Angiogenesis Inhibition Assay provides an efficient system for the rapid and accurate identification of factors that inhibit tube formation of endothelial cells. The µ-Angiogenesis slide, with enhanced optical capabilities in a microscale format, allows for real time and continuous monitoring of tube formation in a simplifed and reproducible manner.

      Storage and Shipping Information
Storage Conditions1. Millicell® µ-Angiogenesis Slides (Part No. CS203030) should be stored at room temperature until ready to use.
      2. ECMatrix™ Gel Solution (Part No. 90060) should be stored at -20ºC until ready to use. Upon thawing, store at 2-8ºC for up to one week. Do not refreeze after thawing as this may affect efficacy.
      3. Calcein AM (Part No. CS202541) is provided as 50 µg lyophilized powder. To prepare 1 mM Calcein AM Stock Solution, briefly centrifuge before opening tube and then add 50 µL sterile DMSO (not provided). Vortex vigorously. Centrifuge briefly to collect solution. Unused Calcein AM solution may be stored in aliquots at -20ºC for up to 1 month.
      4. D, L-Sulforaphane (Part No. CS203032) is provided as 1 mg lyophilized powder. To prepare a 50 mM Stock Solution, dissolve powder in 112 µL sterile DMSO. Store reconstituted D, L-Sulforaphane solution at -20ºC for up to 3 months. Avoid repeated freeze/thaw cycles.
