      Background InformationProtein AF-9 (UniProt: P42568; also known as ALL1-fused gene from chromosome 9 protein, Myeloid/lymphoid or mixed-lineage leukemia translocated to chromosome 3 protein, MLLT3, YEATS domain-containing protein 3) is encoded by the MLLT3 (also known as AF9, YEATS3) gene (Gene ID: 4300) in human. AF9 is a chromatin reader component of the super elongation complex (SEC), which is shown to be essential to increase the catalytic rate of RNA polymerase II transcription by suppressing transient pausing by the polymerase at multiple sites along the DNA. It specifically recognizes and binds acylated histone H3, with a marked preference for histone H3 that is crotonylated. It recognizes and binds histone H3 crotonylated at lysine 9, and with slightly lower affinity histone H3 crotonylated at lysine 18. This binding is achieved through its YEATS domain (aa 8-112). It is also reported to recognize and bind histone H3 acetylated at lysine 9, but with lower affinity than crotonylated histone H3. Mutations of MLLT3 gene have been implicated in neurodevelopmental diseases, including epilepsy and ataxia. A chromosomal aberration involving MLLT3 has also been is associated with acute leukemias.
