      Background InformationZinc finger protein castor homolog 1 (UniProt Q9CWL2; also known as Castor-related protein, Putative survival-related protein, Zinc finger protein 693) is encoded by the Casz1 (also known as Cst, D4Ertd432e, Kiaa3026, Srg, Znf693) gene (Gene ID 69742) in murine species. Originally identified as a Drosophila caster gene product involved in neural fate determination, mammalian CASZ1 protein plays a critical role during the development of a variety of different cell and tissue types, including brain and heart. CASZ1 directly targets and promotes EGFL7 transcription in endothelial cells (ECs), thereby stimulates EGFL7/RhoA-mediated vessel assembly and morphogenesis. Both human and murine express two spliced CASZ1 isoforms/variants (CASZ1a/CASZ1V1 and CASZ1b/CASZ1V2; also known as hCASZ11 and hCASZ5 in human), with the larger isoform containing eight C2H2-type zinc fingers and the short isoform containing only the first four zinc fingers. Loss of CASZ1 expression is found in neuroblastoma tumors of patients with a poor prognosis, while restoration of either CASZ1a/CASZ1V1 or CASZ1b/CASZ1V2 suppresses neuroblastoma tumor growth in cultures. In addition, CASZ1b/CASZ1V2 is reported to bind histones and interact with NuRD (nucleosome-remodeling deacetylase) complex, as well as DNA repair proteins. Disruption of either H3/DNA repair protein or NuRD binding to CASZ1b/CASZ1V2 significantly decreased its transcriptional activity.
