      Background InformationTranscription factor COE3 (UniProt: O08791; also known as Early B-cell factor 3, EBF-3, Olf-1/EBF-like 2, O/E-2, OE-2, EBF-3) is encoded by the Ebf3 (also known as Coe3) gene (Gene ID: 13593) in murine species. EBFs are a family of highly conserved DNA-binding transcription factors with an atypical zinc-finger and helix-loop-helix motif. All members of the EBF family (EBF-2, EBF-3, and EBF-4) possess a highly conserved DNA-binding domain that is distinct from that of other known DNA-binding proteins. These proteins serve diverse functions in the development of multiple lineages, including neurons, B cells, and adipocytes. EBF-3 is highly expressed in adult olfactory epithelium. In the developing embryo, it is expressed in epithalamus, hypothalamus, brainstem, ventricular zones of mesencephalon, and in rhombencephalon. It is highly expressed in developing retina. EBF-3 recognizes variations of the palindromic sequence 5'-ATTCCCNNGGGAATT-3'. EBF-3 is reported to act as a putative tumor suppressor gene in acute myeloid leukemia (AML) and other cancers. Silencing of the EBF3 locus has been observed in brain, colorectal, breast, liver, and bone tumor cell lines. Its reactivation has been reported following treatment with 5-aza-2 -deoxycytidine and Trichostatin A (TSA) and overexpression of EBF-3 is shown to result in cell cycle arrest and apoptosis. EBF3 promoter is reported to be hypermethylated in AML and several cancer cell lines, including HL-60, U937, and K562 cells. (Ref.: Tao, YF., et al. (2015). J. Exp. Clin.
