      Application CommentsFor immunoblotting the antibody should be diluted in PBS, 5% BSA, 0.04% Tween®-20 detergent for 1 h at room temperature. Specificity for total paxillin was confirmed by comparison to a phospho-specific anti-paxillin antibody (see application data). Antibody should be titrated for optimal results in individual systems.

      DescriptionProtein A purified mouse monoclonal antibody. Recognizes the ~68 kDa paxillin protein.
      BackgroundPaxillin is a focal adhesion protein that is involved in the formation of focal adhesions during cell adhesion and migration. The native protein contains LD motifs, LIM domains, and SH3-/SH2-binding domains that facilitate a variety of protein-protein interactions with kinases, GTPase-activating proteins, and cytoskeletal proteins. Phosphorylation of paxillin at Ser178 by JNK1 and cdc2 occurs in response to growth factor activation and fibronectin interaction. Mutation of Ser178 to alanine results in a decrease in the migration of keratinocytes and epithelial cells, suggesting that phosphorylation at this site is an important factor in cell migration.
