      Background InformationHelicase SKI2W (UniProt Q15477; also known as Helicase-like protein, HLP, Ski2) is encoded by the SKIV2L (also known as DDX13, SKI2W, SKIV2, THES2, W) gene (Gene ID 6499) in human. Ski2w is a mammalian homologue of yeast ski2 protein proposed to be involved in RNA processing. The presence of a DEVH-box motif (a.a. 423-426) and its in vitro ATPase/GTPase activity both point to Ski2w being an RNA helicase, although such biological activity has not been demonstrated in vivo. In addition, human Ski2w contains two consecutive leucine zipper motifs commonly involved in protein-protein interaction, as well as an RGD motif (a.a. 238-240) that can potentially interact with cell adhesion molecules. Ski2w is ~140 kDa in size and present in the nucleolus and in the cytoplasm. Ski2w is reported to be associated with ribosomes in an mRNA-independent manner, suggesting a role in protein synthesis regulation. SKIV2L gene mutations have been identified in patients with Tricohepatoenteric syndrome (THES) characterized by intrauterine growth retardation, severe diarrhea in infancy, facial dysmorphism, immunodeficiency, and hair abnormalities, mostly trichorrhexis nodosa.
