      Background InformationSignal recognition particle receptor subunit alpha (UniProt P08240; also known as Docking protein alpha, DP-alpha, SR-alpha, SRalpha) is encoded by the SRPR gene (Gene ID 6734) in human. Secretory proteins typically possess N-terminal hydrophobic signal sequences that direct them to the endoplasmic reticulum (ER) membrane, where they are translocated into the ER lumen for subsequent vesicular transport. Signal recognition particle (SRP) delivers target nascent secretory proteins, while they are still being synthesized, together with the associated ribosome to the ER protein translocon (Sec61 complex) via the interaction with SRP receptor (SR) on the ER membrane. SR is a heterodimer composed of a 70 kDa peripheral membrane protein (SR alpha subunit or SRα) and a 30 kDa singlel-trans membrane protein (SR beta subunit or SRβ). SRα contains an N-terminal SRX domain that mediates its association with SRβ and a C-terminal region which has high homology to the NG domain of SRP54, including a functional GTPase domain. SRβ also possesses a GTPase domain, which interacts with SRα SRX domain only when SRα is in the GTP bound state. SRα forms the major interaction interface with SRP.
