      Background InformationThe Thomsen-nouveau (Tn) monosaccharide is a tumor-associated carbohydrate antigen (TACA) that has been shown to be overexpressed in several carcinomas, including, but not limited to, breast, ovarian, and colon cancers. It is not detected in normal non-cancerous tissues. The Tn antigen (GalNAc) is an O-linked mucin TACA. Mucins are highly glycosylated proteins in epithelial cells. Transmembrane mucins are known to contribute to transmit growth and survival signals to the interior of cells. In many epithelial surfaces, mucins shield and protect cells from infections by pathogens. The Tn antigen arises from a blockage in the normal O-glycosylation pathway in which glycans are extended from the common precursor GalNAcα1-O-Ser/Thr. The precursor is generated in the Golgi apparatus on newly synthesized glycoproteins by a family of polypeptide α-N-acetylgalactosaminyltransferases (ppGalNAcTs) and then extended to the common core 1 O-glycan Galβ1-3GalNAcα1-O-Ser/Thr (T antigen) by a single enzyme termed the T-synthase (core 1 β3-galactosyltransferase or C1GalT).
