      DescriptionRabbit polyclonal antibody supplied as undiluted serum. Recognizes the ~14 kDa p14ARF protein.
      BackgroundThe human p14ARF protein (murine homologue is p19ARF) results from the translation of an alternative reading frame of the CDKN2A (p16INK4) gene. The α transcript gives rise to the p16 gene product while the β transcript gives rise to the p14ARF gene product. Although transcriptionally derived from shared exons, there is little amino acid sequence homology between the two proteins. The function of the p14ARF protein is to activate a p53 response and thereby induce cell cycle arrest. p53 in turn negatively regulates p14ARF expression. The mechanism of action for p14ARF is a direct binding of the COOH terminal domain of MDM2 which in turn forms a ternary complex with p53. Interaction with MDM2 results in the blocking of the MDM2 induced degradation of p53. p14ARF transcription is also modulated by the activity of E2F1 expression. Loss of p14ARF correlates with loss of p16 expression and alteration in expression of both proteins is frequently seen in non-small cell lung carcinoma.
