      Application CommentsDoes not cross-react with the non-phosphorylated α subunit of CaM kinase II or the 60 kDa β subunit in either phosphorylation state. In rat hippocampal cells, this antibody labels cell somas and neuropil, while nuclei are only lightly stained by immunofluorescence. This antibody has been reported to work for ELISA. Antibody should be titrated for optimal results in individual systems.

      DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with HL-1 mouse myeloma cells. Recognizes the ~50 kDa α subunit of CaM kinase II phosphorylated on Thr286.
      BackgroundThe Ca2+/calmodulin-dependent kinase II (CaMKII) is a serine-threonine protein kinase which has been shown to phosphorylate a number of neural substrates including synapsin I, nitric oxide synthase, tryptophan and tyrosine hydroxylase, and the inositol 1,4,5-triphosphate receptors (IP3Rs). CaMKII therefore regulates a broad array of functions including the metabolism of carbohydrates, lipids, and amino acids, neurotransmitter synthesis and release, ion channel and cytoskeletal function, and gene expression. CaMKII is a heterooligomer consisting of two distinct but related isoforms of α and β which are abundantly expressed in neurons of the neocortex, hippocampus, amygdala, and basal ganglia. Following a brief exposure to Ca2+, CaMKII can be converted to a Ca2+-independent form via autophosphorylation of threonine-286, suggesting that CaMKII may play an important physiological role in memory formation. Indeed, both mutant mice deficient in CaMKII, and animals expressing an activated calcium-independent form of CaMKII shows deficiencies in hippocampal long-term potentiation (LTP).
